چکیده

The action of peritoneal exudate cells (PEC) from normal and vaccinia virus infected mice on infectious vaccinia virus particles was investigated in vitra. PEC from immune mice showed a significantly higher infectivity titre reduction (virus clearance, VC) than normal cells. This effect could be clearly attributed to the macrophage. Vaccinia virus multiplied in PEC from normal animals while there was no virus propagation in cells from immunized mice. The release of adsorbed or engulfed virus was reduced significantly in PEC from immunized animals. Anti-vaccinia-antibodies seem to activate normal macrophages to increased virus clearance. This stimulating effect was demonstrable only in the IgG fraction of the antiserum. The activity of macrophages from mice injected three times over aperiod of 14 days with vaccinia virus could be entirely blocked with anti-mouse-IgG, while PEC from mice injected one time six days previously were not inhibited.

In vitro stimulation of sensitized lymphocytes by herpes simplex virus and vaccinia virus.

Splenic lymphocytes from rabbits immunized with herpes simplex virus (HSV) were incubated in vitro with ultraviolet light-inactivated HSV, and the degree of lymphocyte transformation was determined by measurement of the incorporation of [(3)H]thymidine into acid-insoluble material. Lymphocytes from immunized rabbits were stimulated as much as 30-fold, whereas lymphocytes from control rabbits we...

Role of early viral surface antigens in cellular immune response to vaccinia virus.

Infection of mice with the vaccinia virus strain WR, Elstree or DIs, a coditional lethal mutant of vaccinia virus, resulted in the generation of vaccinia virus-specific sensitized cytolytic T lymphocytes (CTL). It could be shown by cross-reactivity between the three strains and by inhibition experiments with specific antisera that early vaccinia surface antigens are sufficient for the generatio...

Plaque size reduction as a measure of viral cell-mediated immunity.

This new assay of viral cell-mediated immunity is sensitive, reproducible, and in many ways resembles the in vivo state. The spread of herpes simplex virus between adjacent monolayer cells was inhibited in the presence of spleen cells from guinea pigs sensitized to that virus. This in vitro control of viral growth was quantified by determining plaque size in monolayers to which were added sensi...

Selective increase in lymphocyte interferon response to vaccinia antigen after revaccination.

Viral antigen prepared by heat inactivation of vaccinia virus stimulated production of interferon in association with transformation of sensitized human lymphocytes in vitro. Involvement of a macrophage-lymphocyte interaction in production of interferon stimulated by viral antigen was found in which macrophage greatly augmented the amount of interferon produced by lymphocytes. Reimmunization wi...

Shared determinants between virus-infected and trinitrophenyl-conjugated H-2-identical target cells detected in cell-mediated lympholysis.

Infection of H-2-identical mice with either lymphocytic choriomeningitis (LCM) virus, vaccinia virus, or paramyxo (Sendai) virus resulted in the generation of specifically sensitized cytotoxic T lymphocytes (CTL). CTL generated in vitro against 2,4,6-trinitrophenyl (TNP)-conjugated syngeneic stimulator cells were specifically cytotoxic for TNP-conjugated H-2K (D) region identical targets. Both ...
